Berliinin museosaari on saanut nimensä siitä, että viiden museon arvokkaat kokoelmat todella sijaitsevat saarella - keskustan halki kiemurtelevan Spree-joen syleilyssä. Siellä, missä aikoinaan seisoivat satamarakennukset ja teollisuushallit, kohoavat nykyään 1800- ja 1900-luvun mahtavat rakennukset, jotka tekevät saaresta yhden Berliinin kuuluisimmista maamerkeistä.

Neues Museum tunnetaan esihistoriallisesta ja egyptiläisestä kokoelmastaan, johon kuuluu myös legendaarinen Egyptin kuningattaren Nefertitin rintakuva.
Alte Nationalgalerie keskittyy romantiikan ja impressionistien kauteen, ja sen kokoelma sisältää Saksan tärkeimpiä taideteoksia.
